CMS and high traffic sites: PHP and MySQL

Question

Is there a difference between CMS and hight traffic sites (for example, news portals) in the logic and design and optimization of databases (PHP and MySQL)? I searched php site scalabilityon stackoverflow and memcached is in most. Are there any MySQL optimization methods? (I searched for a book for this problem. I searched in amazon , but I don’t know what is the best choice.) Thanks in advance
