The easiest way to get started with debugging PHP (Drupal)

What is the easiest way to get started with debugging and pushing a specific line in a Drupal installation?

• Generally?
• With eclipse? (how complicated?)
• On Windows, knowing and having access to Visual Studio (how complicated is it with Visual Studio?)

(No payment: free or trial)

Locally on Windows I use the Acquia Drupal WAMP stack. On the Ubuntu / Webmin / LAMP server.
