You checked the parameter in the webconfig file. You have all the necessary settings for ajax.

Have you tried changing TagPrefix to

<%@ Register Assembly="AjaxControlToolkit" Namespace="AjaxControlToolkit" TagPrefix="asp" %>

directive for anything other than asp? Not sure how well more than one build is supported for one TagPrefix.
