Do you have a web application configured as Medium Trust, and does Full Trust be required to build SQLite.NET?

See if this adds a fix to your web.config:

<system.web>
    <securityPolicy>
        <trust level="Full" />
    </securityPolicy>

...
</system.web>

Or, if you already have a trust tag, change the level to Full.
