Can I develop a system using SQL Server Express and then switch to SQL Server (with the same schema) using the same code (with the exception of the connection) during deployment?

Yes. Just be careful when you take a step to make sure that your instance of SQL Express is at the same service pack level (or lower) as your new version of SQL Standard / Developer / Enterprise, because if it is not, your Databases will not be attached ... People can sometimes get confused about this when they make a switch.
