The "focal length" in the calibration matrix is โ€‹โ€‹not actually expressed in pixels. In fact, this is the ratio that relates the sensor size in the real world to the pixel size in the image. It relates the focal length of the lens to the distances in the image using the zoom factor in each dimension. More details in this article .

As Zaphod says, you can get a decent approximation combining a calibration matrix with a known focal length for the lens in mm.

More accurate method: open a window, read the serial number of the CCD, look at interwebs. There are only a few CCD manufacturers, and they all publish their specifications on the Internet. Here is an example.
